I'm trying to deploy a registry file to turn UAC on using SCCM.

I have the registry file but having a hard time to deploy it through SCCM.

I'm using the command line "regedit.exe IMPORT /s UAC.reg.

It worked once and then hasn't worked after resetting the test pcs back to having UAC off.

Thoughts?

Are you sure that regedit.exe uses "IMPORT" parameter?
I believe you mixed regedit.exe with reg.exe. You should try with one of the following command lines:

Regedit.exe /s UAC.reg

OR

Reg.exe IMPORT UAC.reg

SCCM will only run the job once unless you schedule it unless you set to run more than once (and allow it to do so) or re trigger the job with one of the right click tools the following has this :
ConfigMgr Console Extensions

ConfigMGR Client tools: rerun advertisement

deployment scheduling rerun behavior rerun if failed previous attempt
schedule how often to run
use the version control of the program and content update to push the changes to your distribution point
