You need execute the following powershell cmdlet on the client machine

For AllUsers

Add-AppvClientPackage \\server\share\sample.appv | Publish-AppvClientPackage -Global

Per-User

Add-AppvClientPackage \\server\share\sample.appv | Publish-AppvClientPackage

starting with app-V 5.0 beta2 there are powershell cmdlets available for these kinds of tasks. Have a look here: http://apprepack.blogspot.be/2012/07/to-install-app-v-50-betapowershell.html

you can also make a script that will execute the msi (which is created during the sequence)
