Use the Application Isolation Wizard (AIW.EXE) provided in AdminStudio as one of the standalone tools.

Click Start .. AdminStudio .. AdminStudio Tools .. Application Isolation Wizard and follow the prompts

The Wizard only accepts files of .MSI, .MSM or .EXE format, so you would need to compile your ISM to an MSI first.

What criteria did you use to decide to include the isolated file rather than use the existing one? And presumably this is a file which is protected by Windows?

that your trying to install the system file if you install that it may not work for the existing applications properly

Do NOT include "vbscript.dll" in your package.
Remove it and problem solved.

Is the company you work for getting paid for you to do this? As the client, I'd be bloody petrified of the dross that company must be pushing out. Not only do you not have the first clue about packaging, you're not even aware of basic Windows configuration.

Scary...
