We have to defince a approver by manually selecting the Approver from the list of Approvers, However we want to define a default approver for a Help Desk Queue

I've done this in the past before. We were even able to set custom approvers depending on the user submitting the ticket. Take a look at the custom rules. Give it a shot and post your results. We'll help if you get stuck.

I don't think you can set a default value, but you could have a rule set the approver once the ticket is saved.

You could create the "Aprovers" label, add the label to the specific person who is the approver and then set the queue to only allow "Approvers" by enabling Restrict Approvers by Label.
